Reconstruction methods in holographic microscopy

This master’s thesis focuses on the image reconstruction from holographic microscope. The first part of the thesis summarizes problems of holography, describes its principle and application particularly in live cell biology. In the second part the description of several methods used for off-axis hologram reconstruction is provided. The last part describes the implementation of the basic step in holographic reconstruction – the elimination of autocorrelation and twin image – and then also the reconstruction of 3D information of the sample.
Holographic module for a light microscopy

The new arrangement of the off-axis holographic module, which is using polarizationactive diffraction grating divides signal into reference and subject wave of an interferometer based on their polarization. However, current design of the module does not have a possibility to tune a length of the optical paths. Thus the inability to tune optical paths leads to a reduced quality of interference structure during observation of biological samples. The current module is only suitable for technical applicating due to this limitation. Possibility of tuning branches is key step in biological applications. Therefore a new computer-controlled module is created in order to enable use for biological samples.
Binocular vision

This masters thesis deals with the physiology of binocular vision people for obtaining two dimensional spatial perception of images with glasses. There are developed theoretical part of the physiology of vision, then writing available methods that are now widespread for obtaining stereoscopic scene. The work also describes shooting dynamic scene using a pair of identical cameras. It focuses mainly on OFF-AXES method and deals with the topic of the settings stereoscopic base. There is also realized a practical part, which works with the capture dynamic scenes using video cameras with a automatic control of cameras distance. Automatic control is performed using the parallel port and a stepper motor. For this purpose it is designed to work in hardware support and a program for mutual communication. Filmed a short movie was screened to a group and those observers evaluated the parameters that were specified in the created poll.
Application for Visualization of 3D Objects for Stereoscopic Vision

This thesis is objected to problem, how to create a stereoscopic projection and stereoscopic picture on screen, what has influence on it and how to improve it for nextime. There are described several, generally used, screening technologies and also libraries, in theoretical matter, used in designing, testing and implementation of this thesis. Last part considers about designing, iplementating and testing application of DigitalSpot light system from Robe lighting LTD.
